About this compound

For In Vitro Research Use Only – Not for Human or Veterinary Application

Read this first: racemate vs. AF710B

This product is the racemate, a 1:1 mixture of two mirror-image forms. The primary literature calls the racemate AF710. Separating it gives two enantiomers, AF710A and AF710B. Almost everything published under the name "AF710B" (later ANAVEX3-71) describes the single purified enantiomer, AF710B. The 2016 paper that characterized it reports strong enantioselectivity: AF710A was inactive in its rat model, and its tables list AF710A as an M1 muscarinic antagonist. We found no published pharmacology on the racemate itself.

Identity

1-(2,8-dimethyl-1-thia-3,8-diazaspiro[4.5]decan-3-yl)-3-(1H-indol-3-yl)propan-1-one. C20H27N3OS, 357.5 g/mol. PubChem CID 58516166 for the structure without stereochemistry, and 46846427 and 46846426 for the single enantiomers. Two CAS numbers (1235733-73-9 and 1235733-77-3) sit on the same PubChem record, so we don't attach either to this product with certainty.

Where it comes from

Abraham Fisher's group at the Israel Institute for Biological Research made it, as part of a line of muscarinic agonists (AF102B, AF267B, AF292). It is covered by US patent 8,673,931 (priority 2009). Anavex Life Sciences later took it into development as ANAVEX3-71. A Phase 1 study has been completed and a Phase 2 study is registered.

Mechanism (of the AF710B enantiomer)

AF710B is described as an allosteric M1 muscarinic agonist that is also a sigma-1 receptor agonist. At very low concentrations it strengthened carbachol binding and downstream ERK1/2 and CREB signalling at M1, at a site distinct from acetylcholine's. Some of its effects were blocked by the sigma-1 antagonist NE-100, or by sigma-1 knockdown in cultured neurons.

Research on the enantiomer

Eight PubMed records, concentrated in three groups: Fisher (IIBR), Cuello (McGill) and Bezprozvanny. The models are an M1-antagonist passive-avoidance model in rats, female 3xTg-AD mice, McGill-R-Thy1-APP rats at pre-plaque and post-plaque stages, and dendritic-spine stability in neurons from PS1-KI and APP-KI mice.

This lot

Lot PCS-AF7-0626O, tested as AF710B (racemic): 99.3% by quantitative ¹H NMR at NuMega Resonance Labs. This is chemical purity. It doesn't measure the ratio of the two enantiomers. 50 capsules of 15 mg per bottle.
