Fasoracetam (NS-105)
Fasoracetam has had two research lives. Nippon Shinyaku in Kyoto developed it in the 1990s as NS-105, and a 2018 paper says it went through Phase I–III trials in vascular dementia without approval. Twenty years later Aevi Genomic Medicine revived it (NFC-1 / AEVI-001) for a genotype-selected programme in people with variants in metabotropic glutamate receptor (mGluR) network genes. Nobias Therapeutics has since run a Phase 2 study under the code NB-001.
Structure
It is a pyroglutamide, (5R)-5-(piperidine-1-carbonyl)pyrrolidin-2-one. It's built on 5-oxo-D-proline, unlike the 2-oxopyrrolidin-1-yl-acetamide racetams, and it is a single (R) enantiomer.

- Monohydrate: C10H18N2O3, 214.26 g/mol, PubChem CID 10198262
CAS 110958-19-5 is used inconsistently. Nippon Shinyaku's papers attach it to the monohydrate, while PubChem attaches it to the anhydrous form and gives the monohydrate 2055646-53-0. Solid-state work found two hydrates and one anhydrous form, and hydrate form I melts at only 57 °C.
What the mGluR label rests on
In rat cortical membranes and cultured neurons, fasoracetam moved cAMP in both directions. It inhibited forskolin-stimulated cAMP through Gi/Go proteins, and enhanced it when those were blocked. An mGluR antagonist blocked both effects. Antagonist and antisense work tied the inhibitory side to group II/III mGluRs and the facilitatory side to group I. This is indirect functional evidence from one lab. No binding or structural study confirms direct mGluR binding, and the 2018 authors note that other mechanisms may exist. Separate studies reported more GABA-B receptors in rat cortex after repeated dosing, and cholinergic effects in rat amnesia models.
Other research
Radiolabel pharmacokinetics in rats, dogs and monkeys, rodent memory-disruption and forced-swim models, and genetics papers on mGluR-network copy-number variants.
Pack
50 capsules of 20 mg per bottle (Peak Ops). No certificate of analysis is posted for fasoracetam yet, so we quote no purity figure and don't state whether the lot is the hydrate or the anhydrous form.
