NSI-189: found by screening for an effect
Most compounds on our catalog were designed against a known receptor. NSI-189 went the other way. Neuralstem, Inc. picked it out of a phenotypic screen because it stimulated neurogenesis in human hippocampus-derived neural stem cells, and later in mouse hippocampus. Years on, its molecular target still hasn't been identified. It has since received the INN amdiglurax.
Chemistry: (4-benzylpiperazin-1-yl)-[2-(3-methylbutylamino)pyridin-3-yl]methanone, a benzylpiperazine-aminopyridine. The free base is C22H30N4O, 366.5 g/mol, CAS 1270138-40-3, PubChem CID 50922681. We supply NSI-189 as the free base (the form described above).
What's been observed
Its activity is described as independent of monoamine reuptake. In cell and animal studies, researchers have linked it to TrkB and Akt activation, higher mitochondrial function and AMPK activity, and higher BDNF and SCF expression. These are downstream readouts, not a binding site.
An unusually wide range of models
| Model | System |
|---|---|
| Hippocampal neurogenesis | Human neural stem cells; mice |
| Angelman syndrome | Mouse model (hippocampal LTP) |
| Ischemic stroke | Rat middle cerebral artery occlusion |
| Cranial irradiation | Rats |
| Diabetic neuropathy | Type 1 and type 2 mouse models; Zucker diabetic fatty rats |
| Oxygen-glucose deprivation | Hippocampal cell cultures |
Most of this work was done by the developer or with it. Neuralstem ran Phase 1 and Phase 2 studies. Newer neurogenesis compounds are now benchmarked against it.
