PRL-8-53: the whole record
Few compounds this widely discussed have less published about them. The complete record for PRL-8-53 is two documents, both from its inventor, Nikolaus R. Hansl:
- US patent 3,870,715 (filed 1973, granted 1975), "Substituted amino ethyl meta benzoic acid esters". It describes rat maze and shock-avoidance tests, isolated-ileum spasmolytic tests, and rodent toxicology for the compound series. It calls them "cholinergic and spasmolytic agents".
- One 1978 paper in Psychopharmacology (Hansl & Mead), a small double-blind study.
A PubMed search for PRL-8-53 returns that one paper. We found no independent replication, no receptor-binding study and no later research.

Mechanism
Unknown. The patent's "cholinergic" label rests on a gut-muscle (ileum) assay. Mechanisms you may see elsewhere, such as dopamine or serotonin effects, have no primary source.
